^Only two things:

1. Sell 'cuda;
2. Buy Charger R/T.

I remember having 'cuda as dragster about year ago, and after one whole day of (unsuccessful) performance tuning I gave up and sold it.

Anyway, if you are such a fan of Hemicuda, try increasing gear ratios (drag) or stiffening suspensions and increasing height (wheelie). Won't help much, but you can try.

New GT500 is good. Didn't try the old one, but I guess EA made it too weak for anything better than 9 seconds quarter.

Which makes me think I should try it...

Any AWD car is good for speed. The best is Murcielago LP640 IMO.

As for tuning, I can't really say what is best for you because I don't know your driving style. You should test it on the track and tune it how you like it.

OK now, this is a setup for Corvette C6 but you can use on the 2006 GT500. I am using this setup and my best time is 5.95 s.

Install stage 4 suspension before tuning, then after you have adjusted everything, drop it to stage 2.
Stage 4 Corvette C6 (drop suspension to stage 2 after tuning):

Suspension:
Shocks, Springs, Rebound: 10 clicks right
Ride Height: 10 clicks right
Front Anti-Roll Bar: 10 clicks right
Rear Anti-Roll Bar: 10 clicks left
Front Tire Pressure: 10 clicks right
Rear Tire Pressure: 10 clicks left
Toe, Camber: 1 click right
Caster: 1 click left
Steering: 10 clicks right

Engine:
Start, End Boost: 10 clicks right
Nitrous Pressure, Flow Rate: 10 clicks right

Drivetrain:
1st: 10 clicks left
2nd: center (10 clicks right is also useful)
3rd-6th: 10 clicks right
Final: 10 clicks right

I once tried to tune a Cuda for drag but couldn't get any grip in the rear tires no matter what... Gave up on it shortly after. Ironically its sister car (Dodge Challenger) is one of the best dragsters in the game.
